Cell-level dosimetry and biologic response modeling of heterogeneously distributed radionuclides: a step forward.

چکیده

Historically, clinical response to internal radionuclides has been related to the macroscopic (i.e., whole-tumor or whole-organ) absorbed dose, implicitly assuming uniform distributions of activity and energy deposition (1,2). However, the actual biologic response among cells within a tumor or organ can vary substantially, depending on spatial nonuniformities of these distributions at the multicellular, cellular, and subcellular levels (3–9). The macroscopic, or mean, absorbed dose thus may not be a reliable descriptor of the biologic effect of internally deposited

Cellular dosimetry of beta emitting radionuclides-antibody conjugates for radioimmunotherapy

Introduction: The choice of optimal radionuclides for radioimmunotherapy depends on several factors, especially the radionuclide and antibody. The dosimetric characteristics of a non-internalizing and an internalizing monoclonal antibody (MAb) labeled with beta emitting radionuclides were investigated. Methods: Using Geant4-DNA Monte Carlo simulation, we ...

Risks to normal tissues from radionuclide therapy.

The development of radionuclide therapies during the past few decades provides a growing body of data on radiobiologic effects, including normal tissue toxicities and antitumor efficacy. Information on normal tissue toxicity from radionuclides is more limited than that from external beam radiation and appears to be more variable. Much of the increased variability is attributed to heterogeneous ...

Influence of nonuniform activity distribution on cellular dosimetry.

Dosimetry of nonuniformly distributed 210Po,67Cu, and 123I was evaluated for a single cell, modeled as a homogeneous unity-density sphere with a radius of 7.7 mum, which is typical of a B-cell lymphoma cell. A Visual Basic program was used to calculate S factors and radial absorbed dose-rate profiles for these radionuclides, taking into account their individual radiation energy spectra. The res...

Absorbed Dose Assessment from Short-Lived Radionuclides of Radon (222Rn) Decay Chain in Lung Tissue: A Monte Carlo Study

Introduction: Internal exposure to radon gas progeny can lead to serious biologic damages to the lung tissue. The aim of this study was to evaluate the absorbed dose by lung tissue due to the exposure from short-lived radioactive products of radon (222Rn) decay using Monte Carlo simulation. Material and Methods: A lung equivalent p...

Log normal distribution of cellular uptake of radioactivity.

UNLABELLED It is widely recognized that radiopharmaceuticals are generally distributed nonuniformly in tissues. Such nonuniformities are observed over the entire range of spatial levels, ranging from organ to subcellular levels. The implications of nonuniform distributions of radioactivity for dosimetry, and ultimately for the biologic response of tissues containing radioactivity, have been inv...
